The cluster of differentiation (CD) system is commonly used as cell markers in immunophynotyping. Different kinds of cells in the immune system can be identified through the surface CD molecules which associating with the immune function of the cell. There are more than 320 CD unique clusters and subclusters have been identified. Some of the CD molecules serve as receptors or ligands important to the cell through initiating a signal cascade which then alter the behavior of the cell. Some CD proteins do not take part in cell signal process but have other functions such as cell adhesion. CD120a (cluste of differentiation 120a), also known as TNFR1 / TNFRSF1A, is a member of CD family, tumor necrosis factor receptor superfamily. CD120a is one of the most primary receptors for the tumor necrosis factor-alpha. It has been shown to be localized to both plasma membrane lipid rafts and the trans golgi complex with the help of the death domain (DD). CD120a can activate the transcription factor NF-KB, mediate apoptosis, and regulate inflammation processes.
 
【检验原理】
本试剂盒采用双抗体夹心ELISA法。酶标微孔板上预包被有抗小鼠TNFRSF1A抗体，当加入标准品和待测样本时，标准品和待测样本中的小鼠TNFRSF1A会和该抗体结合。洗板后，游离的其他成分被洗去，加入辣根过氧化物酶标记的的抗小鼠TNFRSF1A抗体可与被酶标板捕获的小鼠TNFRSF1A结合，形成“包被抗小鼠TNFRSF1A抗体+小鼠TNFRSF1A+抗小鼠TNFRSF1A抗体-HRP”免疫复合物。当加入TMB显色底物后，复合物上连接的HRP催化底物反应，生成蓝色产物，终止反应后变为黄色。在450nm处测定OD值，小鼠TNFRSF1A浓度与OD450值之间呈正比，可通过绘制标准曲线求出标本中小鼠TNFRSF1A的浓度。
