Gp130, the common signal transducing receptor component shared by the functional receptor complexes of the IL-6 family of cytokines, belongs to the class I cytokine receptor family. Binding of IL-6 (IL-11) to either the membrane-anchored or soluble IL-6 R (IL-11 R) initiates the association of IL-6 R (IL-11 R) with gp130 which then undergoes homo-dimerization and signal transduction. With other IL-6 family cytokines, such as LIF and OSM, signal transduction is triggered by the hetero-dimerization of gp130 and LIF R or OSM R.Gp130 is expressed in all organs examined. Soluble gp130, which apparently arises either from proteolytic cleavage of the membrane-bound receptor or from alternative splicing, has been detected in human serum. The in vivo functions of soluble gp130 are not clearly understood. Inin vitro experiments, natural or recombinant soluble gp130 has been shown to have inhibitory effects on OSM and CNTF activities.
 
【检验原理】
本试剂盒采用双抗体夹心ELISA法。酶标微孔板上预包被有抗小鼠gp130抗体，当加入标准品和待测样本时，标准品和待测样本中的小鼠gp130会和该抗体结合。洗板后，游离的其他成分被洗去，加入辣根过氧化物酶标记的的抗小鼠gp130抗体可与被酶标板捕获的小鼠gp130结合，形成“包被抗小鼠gp130抗体+小鼠gp130+抗小鼠gp130抗体-HRP”免疫复合物。当加入TMB显色底物后，复合物上连接的HRP催化底物反应，生成蓝色产物，终止反应后变为黄色。在450nm处测定OD值，小鼠gp130浓度与OD450值之间呈正比，可通过绘制标准曲线求出标本中小鼠gp130的浓度。
