Interleukin 19 (IL-19) is a member of the IL-10 family of cytokines . Mouse IL-19 is synthesized as a 176 amino acid (aa) precursor that contains a 24 aa signal sequence and a 152 aa mature region . Based on human studies, it is expected to be secreted as a glycosylated monomer, 35 - 45 kDa in size. IL-19 is unusual in that it contains seven amphipathic helices. Mature mouse IL-19 shares 69% aa sequence identity with the mature human IL-19, and 85% and 68% aa identity to unpublished Genbank sequences for rat and canine IL-19, respectively. Although mouse IL-19 is active on human cells, human IL-19 is not active on mouse cells. IL-19 expression is limited to activated keratinocytes and monocytes, with a possible contribution from B cells. IL-19 binds a receptor complex consisting of the IL-20 receptor alpha (also known as IL-20 R1) and the IL-20 receptor beta (IL-20 R2). 
 
【检验原理】
本试剂盒采用双抗体夹心ELISA法。酶标微孔板上预包被有抗小鼠IL-19抗体，当加入标准品和待测样本时，重组或天然的小鼠IL-19会和该抗体结合。洗板后，游离的其他成分被洗去，加入生物素化的抗小鼠IL-19抗体可与被酶标板捕获的小鼠IL-19结合。再次洗板后，游离的其他成分被洗去，加入辣根过氧化物酶标记的链霉亲和素，生物素和链霉亲和素特异性结合，形成“包被抗小鼠IL-19抗体+小鼠IL-19+生物素化抗小鼠IL-19抗体+链霉亲和素-HRP”免疫复合物。当加入TMB显色底物后，复合物上连接的HRP催化底物反应，生成蓝色产物，终止反应后变为黄色。在450nm处测定OD值，小鼠IL-19浓度与OD450值之间呈正比，可通过绘制标准曲线求出标本中小鼠IL-19的浓度。
