Interleukin10 Receptor beta (IL10 Rβ), also known as IL10 R2 and CRF24, is a 60 kDa transmembrane glycoprotein that functions as a coreceptor for several class 2 cytokines including Interleukins10, 22, 26, 28A/ IFNλ2, 28B/ IFNλ3,
and 29/IFNλ . IL10 Rβ associates with ligandspecific receptor subunits to form signaling receptor complexes, e.g. IL10 Rα for IL10, IL20 Rα for IL26, IL22 Rα for IL22 , and IL28 Rα for IL28A, IL28B,and IL29. IL10 Rβ is widely expressed, while the associated receptor subunits exhibit differential expression patterns. The ligandspecific subunits are responsible for the divergent functions of these cytokines, encompassing immune suppression, promotion or inhibition of inflammation, mucosal defense, antiviral immunity, and hematopoiesis. IL10 Rβ deficient mice lack responsiveness to each of those cytokines. IL10 Rβ contributes to ligand binding, but effective signaling is onlytriggered in the presence of the ligandspecificsubunit. In the case of IL10, a cytokine dimer binds to two IL10 Rα/IL10R1 chains, resulting in recruitment of two IL10 Rβ/IL10R2 chains. 
 
【检验原理】
本试剂盒采用双抗体夹心ELISA法。酶标微孔板上预包被有抗人IL-10Rβ抗体，当加入标准品和待测样本时，标准品和待测样本中的人IL-10Rβ会和该抗体结合。洗板后，游离的其他成分被洗去，加入辣根过氧化物酶标记的的抗人IL-10Rβ抗体可与被酶标板捕获的人IL-10Rβ结合，形成“包被抗人IL-10Rβ抗体+人IL-10Rβ+抗人IL-10Rβ抗体-HRP”免疫复合物。当加入TMB显色底物后，复合物上连接的HRP催化底物反应，生成蓝色产物，终止反应后变为黄色。在450nm处测定OD值，人IL-10Rβ浓度与OD450值之间呈正比，可通过绘制标准曲线求出标本中人IL-10Rβ的浓度。
