Human Ficolin1 (fibrinogen/collagenlike), also called Mficolin, is a member of the ficolin family of secreted pattern recognition proteins in the lectin complement activation pathway. Ficolin1 is expressed by monocytes, neutrophils and type II alveolar epithelial cells. It is proposed to be a locallyacting lectin released in a regulated manner. Ficolin1 is not found in plasma, but is detected on the surface of circulating monocytes. The 35 kDa, 326 amino acid (aa) human Ficolin1 contains a 28 aa signal sequence, an Nterminal collagen domain and a Cterminal Fibrinogenlike (FBG) domain that includes a calcium binding site and one potential Nglycosylation site. Both the collagen and FBG domains mediate trimer formation. Like Ficolin2, larger homomultimers of Ficolin1 exist and are likely formed by disulfide bonds at the Nterminus. Sizes corresponding to 12 or 18 subunit oligomers are reported . The FBG domain of Ficolin1 binds microbial ligands that contain acetylated compound. Ligands identified include Nacetyl glucosamine, Nacetyl galactosamine and sialylNacetyllactosamine. Mature human Ficolin1 shares 76%, 61%, 61%, 76% and 81% aa identity with mouse Ficolin2 , and mouse, rat, canine and porcine Ficolin1,
 
【检验原理】
本试剂盒采用双抗体夹心ELISA法。酶标微孔板上预包被有抗人ADAM15抗体，当加入标准品和待测样本时，标准品和待测样本中的人ADAM15会和该抗体结合。洗板后，游离的其他成分被洗去，加入辣根过氧化物酶标记的的抗人ADAM15抗体可与被酶标板捕获的人ADAM15结合，形成“包被抗人ADAM15抗体+人ADAM15+抗人ADAM15抗体-HRP”免疫复合物。当加入TMB显色底物后，复合物上连接的HRP催化底物反应，生成蓝色产物，终止反应后变为黄色。在450nm处测定OD值，人ADAM15浓度与OD450值之间呈正比，可通过绘制标准曲线求出标本中人ADAM15的浓度。
