C-Kit is a type 3 transmembrane receptor for MGF (mast cell growth factor, also known as stem cell factor). c-Kit contains 5 Ig-like C2-type (immunoglobulin-like) domains.and 1 protein kinase domain. It belongs to the protein kinase superfamily, tyr protein kinase family and CSF-1/PDGF receptor subfamily. C-Kit contains 5 Ig-like C2-type (immunoglobulin-like) domains and 1 protein kinase domain. C-Kit has a tyrosine-protein kinase activity. Binding of the ligands leads to the autophosphorylation of KIT and its association with substrates such as phosphatidylinositol 3-kinase. Antibodies to c-Kit are widely used in immunohistochemistry to help distinguish particular types of tumour in histological tissue sections. Mutations in c-Kit gene are associated with gastrointestinal stromal tumors, mast cell disease, acute myelogenous lukemia, and piebaldism. Defects in KIT are a cause of acute myelogenous leukemia (AML). AML is a malignant disease in which hematopoietic precursors are arrested in an early stage of development. Note=Somatic mutations that lead to constitutive activation of KIT are detected in AML patients.

【检验原理】
本试剂盒采用双抗体夹心ELISA法。酶标微孔板上预包被有抗人C-Kit抗体，当加入标准品和待测样本时，标准品和待测样本中的人C-Kit会和该抗体结合。洗板后，游离的其他成分被洗去，加入辣根过氧化物酶标记的的抗人C-Kit抗体可与被酶标板捕获的人C-Kit结合，形成“包被抗人C-Kit抗体+人C-Kit+抗人C-Kit抗体-HRP”免疫复合物。当加入TMB显色底物后，复合物上连接的HRP催化底物反应，生成蓝色产物，终止反应后变为黄色。在450nm处测定OD值，人C-Kit浓度与OD450值之间呈正比，可通过绘制标准曲线求出标本中人C-Kit的浓度。
