Marapsin, pancreasin, and channelactivating protease 2 (CAP2) are different names for the same serine protease that is expressed strongly in the pancreas . The protein is synthesized with a signal peptide (amino acid residues 122), a pro peptide (residues 2334) and a mature chain (residues 35290) corresponding to the serine protease domain. The fulllength protein was expressed and the secreted protein purified. The Nterminal sequencing results indicate that the purified protein is a disulfide bondlinked dimer formed between the pro peptide and the mature chain.

【检验原理】
本试剂盒采用双抗体夹心ELISA法。酶标微孔板上预包被有抗人Pancreasin抗体，当加入标准品、待测样本和辣根过氧化物酶标记的抗人Pancreasin抗体后，经过孵育，标准品和样品中含有的人Pancreasin可同时与酶标板的包被抗体和辣根过氧化物酶标记的检测抗体相结合。洗板后，游离的其他成分被洗去，形成“包被抗人Pancreasin抗体+人Pancreasin+抗人Pancreasin抗体-HRP”免疫复合物。当加入TMB显色底物后，复合物上连接的HRP催化底物反应，生成蓝色产物，终止反应后变为黄色。在450nm处测定OD值，人Pancreasin浓度与OD₄₅₀值之间呈正比，可通过绘制标准曲线求出标本中人Pancreasin的浓度。
